Role and Contribution
The preclinical participation of the group will consist in developing a technique to visualise in mice by positron emission tomography the fate of viable dendritic cells after injection through various routes (intra-tumoural, subcutaneous, intravenous, intraperitoneal, microsurgically intranodal). The idea will be to gene mark the dendritic cells with an adenovirus encoding thymidine kinase. Such enzyme in viable cells will phosphorylate a gancyclovir analogue marked that is retained inside the TK+ cells. Once this technique is in place the goal is to use it to monitor migration of dendritic cells under the influence of IFN and PolyI:C in mice. The advantage is that only the cells with a preserved membrane will retain the contrast, and thus it will be possible to monitor cell viability and migration at the same time.
